It is produced endogenously in humans through enzymatic pathways. H2S is associated with the proliferation or inhibition of diseases such as cancer, Alzheimer's, diabetes, and cardiovascular diseases at unknown H2S concentrations. Biochemical and physiological studies of H2S has been realized using compounds that mimic the production of physiological H2S in biological samples for its therapeutic attribution known as H2S donors, but these have shown some disadvantages. Therefore, it is crucial to find a reliable H2S-releasing compound that functions in vitro and in vivo studies with reliable characteristics and controllable properties for better understanding the attribution of H2S in physiological processes, which still unclear. Hemoglobin I from the clam Lucina Pectinata, it was used for the delivery of H2S in vitro to obtain a reliable H2S donor at physiological conditions using spectroscopic techniques. UV-Vis spectroscopic, kinetic experiments of (His)6Tag rHbI(FeIII)-SH2 complex under physiological parameters (pH 7.4, Kreb’s buffer solution, oxygen exposure, temperature 37.0ºC) have shown very slow dissociation rate constants (koff) and long half-life (t1/2). These results demonstrate the slow-released and high affinity of the H2S to the active site of the protein under these conditions, proving that this H2S-delivery protein complex could lead us to the generation of new therapies based on H2S.
