Publication Phase contrast modalities integrated in a portable microscope setup(2024-07-10) Ramos Álamo, María del Carmen; Sierra Gil, Heidy; College of Engineering; Arzuaga, Emmanuel; Vélez Rivera, Bienvenido; Other; George, Dibin M.Several optical microscopy imaging techniques provide the capability to capture quantitative information at high temporal and spatial resolution from live specimens. Usually, high-contrast and quantitative microscopy require chemical staining or tagging, altering physiological features of the sample, and limiting in vitro applications. Alternative noninvasive methods use phase contrast and interferometry to capture quantitative phase images without altering the specimen. However, some of these techniques have complex hardware implementations and high cost that restrict their use to specialized medical centers and facilities. This thesis focuses on developing a portable microscopy system that combines computational models and reduced hardware components to produce high-resolution images and quantitative data. It achieves this by comparing different illumination techniques and deep neural networks (DNN) models. Quantitative phase imaging modalities are adapted in a customized portable phase contrast microscope connected to an LED Matrix Panel and a smartphone. An application was developed to manage the image acquisition and send the collected data to a local server for processing, to enhance the portability of the system. DNN models for reconstructing phase contrast and the optical path difference (OPD) values were explored. The resulting images were compared with those obtained using classical methods, providing a comprehensive understanding of the system’s capabilities.
