A total of 20 male rabbits were assigned to two treatment groups over 8 weeks, including 10 receiving water (control group; T1), and 10 receiving retinyl palmitate as a supplement (experimental group; T2). Both experimental groups received 500 IU of vitamin A in their daily food. The T2 group also received 20,000 IU of vitamin A (A total of 20,500 IU of vitamin A daily). The T1 group received water as a placebo. Semen was collected once per week. Sperms were analyzed using the CASA IVOS II System. Data were analyzed using the Generalized Linear Model Mixed Model. Week, Treatment, and Week and Treatment interacted (p < 0.0001) in total sperm motility (progressive, motile, static, and slow), but not in T1. By the final week, the T2 motility had dropped to an average of 54.88%, with progressive sperm decreasing to averages of 7.43% and slow sperm increasing to 70.43%, while static sperm rose above 45%, when compared with T1. These findings suggest that long term high-dose vitamin A supplementation induces time dependent sperm changes in rabbits, impairing motility and some parameters in sperm morphology.
