The bacterial diversity used to bioremediate gasoline contaminated groundwater in a fluidized bed reactor (FBR) was determined. Isolation and characterization of bacterial populations from the bioremediation system was performed in R2A media. Test included reaction to the Gram coloration, carbon utilization profiles (Biolog®), ability to use gasoline as the sole carbon source and molecular characterization by amplified ribosomal DNA restriction analysis (ARDRA), Box-PCR and 16S rDNA sequence analysis. From a total of 162 distinctive isolates, 75% were gram negative bacilli, 19% gram positive bacilli, 5% gram negative cocci and 1% gram positive cocci. The 162 axenic cultures corresponded to 90 different genetics groups; 53% of which growth in gasoline as sole carbon source. The 86 strains capable of growing in gasoline corresponded to 52 different amplifications patterns in Box-PCR; which are not metabolically related (Biolog® system). In addition, the 16S rDNA of twenty strains were sequenced. The sequences were associated with 3 well-recognized divisions. Phylogenetically, 13 of the sequences are grouped with the alpha, beta and gamma subdivision of the Proteobacteria, one (BW2) with Flexibacter-Cytophaga- Bacteroides group, and six with Firmicutes division (bacterial with High GC content of DNA and Bacillus-Lactobacillus-Streptococcus subdivision). The high degree of microbial diversity in the FBR promoted efficient and stable hydrocarbon removal (>99%), throughout the operation of the system. A better understanding of the biotic behavior will allow to improve and to optimize these units of treatment.
