dc.description.abstract | Analyses of the concentration of the eicosanoid 12-hydroxyeicosatrienoic acid (12- HETE) and heat shock proteins in response to stress conditions were studied in the common Caribbean benthic marine alga Murrayella periclados (C. Agardh) Schmitz, after experimentally exposing the alga to different conditions of temperature and salinity. The eicosanoid concentration in lipid extracts and presence of stress proteins in protein extracts were evaluated in field-collected (wild) and cultured M. periclados. Cultured M. periclados was experimentally exposed to low and high temperature (15 oC and 35 oC) and low and high salinity (10 and 65 ppt) treatments. Control cultures were maintained at 25 oC and 35 ppt. Average values of eicosanoid concentration in field-collected M. periclados ranged from 575 to 750 ng/g, while under culture control conditions they ranged from 304.5 to 325.0 ng/g. Effect of treatments was evaluated immediately (time = 0) and at 10, 30 and 60 minutes after cessation of treatments. A significant increase in eicosanoid concentration was observed 10 minutes after cessation of high temperature and high salinity treatment periods, where average values of eicosanoid concentration ranged from 400 to 510 ng/g. The eicosanoid concentration in samples from low temperature and low salinity treatments was not different from control, and there was no evidence of stress protein expression in these samples. Stress protein 70 and stress protein 25 were the common stress proteins expressed in M. periclados following 30 and 60 minute exposures to high temperature and high salinity and analyzed immediately after treatment (time = 0). The observation of subsequent induction of heat shock proteins in treatments before the increase in 12-HETE concentration, provides an undefined role of this compound in stress response during treatment. However, the eventual increase in 12 HETE, 10 minutes after cessation of treatments, may be is related to the sustained presence of stress protein 25 after 30 and 60 minutes following high salinity and high temperature stress cessation. Agardh) Schmitz, se utiliza como modelo para el análisis de la concentración del eicosanoide 12- ácido hidroxieicosatrieno (12-HETE) y la producción de proteínas de impacto en circunstancias de alta y baja temperatura y salinidad. También se evaluó la concentración de eicosanoides en extracciones de lípidos y la presencia de las proteínas de impacto en extractos de proteínas de M. periclados colectadas en su ambiente natural y en condiciones controladas de cultivo. Experimentalmente se expuso el alga a temperaturas de 15 y 35 oC y a salinidades de 10 y 65 ppt. El control estaba representado por los cultivos de M. periclados mantenidos a 25 oC y 35 ppt. En algas colectadas en su ambiente natural, las concentraciones de eicosanoides fueron de 575 a 750 ng/g, mientras que en las muestras control fueron de 304.5 a 325 ng/g. El efecto del tratamiento se determinó inmediatamente (tiempo = 0) y a 10, 30 y 60 minutos, luego de finalizado cada tratamiento. Se observó un aumento en la concentración de eicosanoides al cabo de 10 minutos luego de concluido los periodos de duración de los tratamientos de alta temperatura y alta salinidad, donde los valores de concentración de eicosanoides fueron de 400 a 510 ng/g. La concentración de eicosanoides en muestras expuestas a baja temperatura y salinidad no fue diferente de las muestras control, tampoco hubo evidencia de la expresión de proteínas de impacto en estas muestras. Las proteínas de impacto 70 y 25 fueron las más comunes en M. periclados en tratamientos de 30 y 60 minutos en alta temperatura y salinidad, inmediatamente culminó el tratamiento (tiempo=0). Esta observación en la inducción de proteínas de impacto en tratamientos antes del aumento en la concentración de 12- HETE, demuestra un rol no definido del compuesto en la respuesta al impacto durante el tratamiento. Sin embargo, un posterior aumento en 12-HETE, a los 10 minutos de culminar los tratamientos, pudiera explicar que se mantenga la presencia de la proteína de impacto 25 a los 30 y 60 minutos de culminar el impacto de alta temperatura y salinidad. Este estudio reconoce por primera vez la expresión de la proteína de impacto 25 en las algas pertenecientes a las florideoficeas. | en_US |
