Genetic expression of HSP70, ATP1A1, NTRK1 and PRLR in the skin of slick hair Holstein cows

dc.description.abstract The skin helps to regulate the core temperature of many organisms, including dairy cattle. Skin glands have been observed in dairy cattle with slick hair genotype which appears to improve their capacity to regulate core temperature. For example, slick-hair cows have increased sweating rates, different sweat and sebaceous gland morphology, and shorter hair. To further study the effects of the slick-hair genotype in cattle, we investigated the expression of four genes in the skin (HSP70, ATP1A1, NTRK1, and PRLR) which have possible effects on thermoregulation in the skin. Skin biopsies were obtained from Holstein cows (slick-hair and wild -type, n=25 per genotype) of the Agricultural Experimental Station, in Lajas, Puerto Rico. RNA was extracted from the skin biopsies and then cDNA was obtained by performing reverse transcriptase reactions. Specific primers were combined with the cDNA to determine the genomic expression of each of the aforementioned genes by using Real-Time PCR. The expression of all four genes was similar (p>0.05) in the skin of slick-hair and wild-type Holstein cows. Additional research is needed to investigate possible differences between slick-hair and wild-type cattle in the expression of these genes in other organs/tissues of the body. en_US
dc.description.abstract La piel ayuda a regular la temperatura corporal en muchos organismos, incluyendo el ganado lechero. Se ha observado diferentes morfologías en glándulas de la piel del ganado lechero que posee el genotipo pelón (“slick”), y que a su vez aparentan mejorar su capacidad para regular la temperatura corporal. Por ejemplo, el ganado pelón, tienen mayor tasa de sudoración, diferentes morfologías de las glándulas sudoríparas y sebáceas y pelo más corto. Para estudiar los efectos del genotipo pelón, se investigó la expresión genética en la piel de vacas pelonas y no pelonas para cuatro genes (HSP70, ATP1A1, NTRK1 y PRLR) que pueden tener un efecto en termorregulación. Se obtuvieron biopsias de vacas Holstein (Pelonas, No Pelonas, n=25 por genotipo) en la Estación Experimental de Lajas, Puerto Rico. Se extrajo RNA de las biopsias de piel y se obtuvo el DNA complementario para determinar la expresión genética de cada uno de los genes utilizando PCR en “tiempo real”. La expresión de estos cuatro genes en la piel de Holstein pelonas y no pelonas fue similar (p>0.05). Por ende, se recomienda investigación adicional para establecer las posibles diferencias entre vacas pelonas y no pelonas para la expresión de dichos genes en otros órganos del cuerpo. en_US
