First integrative study of the diversity and specificity of Metacercariae of <i>Posthodiplostomum</i> Dubois, 1936 from native and introduced fishes in the Caribbean

Abstract
Metacercariae of the genus Posthodiplostomum are often recorded in freshwater fish second intermediate hosts. While the diversity and taxonomy of this genus are receiving increasing attention in molecular phylogenetic studies, available data remain geographically biased. Most molecular studies of Posthodiplostomum and morphologically similar (neascus) worms originate in North America and Europe and Asia (more than 60% of DNA sequences are from USA and Canada), with few data currently available from the Neotropics, where high host diversity suggests high and under-sampled parasite diversity. In this study, we report molecular and morphological data from metacercariae of Posthodiplostomum in fish in Puerto Rico, where only a single species has been previously reported. Partial sequences of cytochrome c oxidase subunit 1 (COI) from metacercariae from Dajaus monticola (native to Puerto Rico) and the introduced fishes Poecilia reticulata, Parachromis managuensis, Lepomis macrochirus and Micropterus salmoides revealed seven genetically distinct specieslevel lineages, of which four were novel. Most species were morphologically distinct and all displayed narrow specificity for fish hosts. The six species of Posthodiplostomum recovered from introduced fishes were not found in native hosts.

Metacercarias del género Posthodiplostomum a menudo infectan peces de agua dulce como segundos huéspedes intermedios. Si bien la diversidad y la taxonomía de este género están recibiendo una atención cada vez mayor en los estudios filogenéticos moleculares, los datos disponibles siguen estando geográficamente sesgados. La mayoría de los estudios moleculares de Posthodiplostomum y gusanos morfológicamente similares (neascus) se originan en América del Norte y Europa (más del 60% proviene sólo de Estados Unidos y Canadá), con pocos datos del Neotrópico, zona que tiene una gran diversidad de hospedadores, lo que supone un mayor número de parásitos por esfuerzo de muestreo. En este estudio reportamos datos moleculares y morfológicos de metacercarias de Posthodiplostomum en peces en Puerto Rico, donde sólo una especie ha sido reportada previamente. Secuencias parciales de la subunidad 1 del citocromo c oxidasa (COI) de metacercarias de Dajaus monticola (nativo de Puerto Rico) y los peces introducidos Poecilia reticulata, Parachromis managuensis, Lepomis macrochirus y Micropterus salmoides revelaron siete linajes genéticamente distintos a nivel de especie, de los cuales cinco son nuevos para la ciencia. Las siete especies eran morfológicamente distintas y mostraban una estrecha especificidad por los peces huéspedes; además, seis especies de Posthodiplostomum se encontraron infectando peces introducidos y sólo una especie en un huésped nativo a la isla.
