The influence of thermic stress on the marine yeast Debaryomyces hansenii

Abstract
Often organisms must acclimate and adjust their metabolism to better adapt to an ever-changing environment. Some environments can be considered to have normalized parameters, while others truly challenge the homeostatic state of many microorganisms, although under both life seems to thrive. This project will attempt to understand these adaptations using the marine yeast Debaryomyces hansenii as a model system. Debaryomyces hansenii is an ascomycete marine yeast initially isolated from a Swedish estuary. It has been shown to tolerate much higher sodium chloride (NaCl) concentrations than those found in seawater. This yeast can grow in the presence of heavy metals as well as in a wide pH range. Moreover, D. hansenii is known to increase population at temperatures ranging from 11oC to 37oC (24oC, optimal), a broader scope than that reported for the most commonly known yeasts. Temperature, in particular, has been known to alter the tolerance response of D. hansenii toward other stressors. For example, in combination with high temperatures (~34C), NaCl stimulated growth beyond the recorded maximum for either of the stressors individually. Also, a higher tolerance against multiple stressors was observed at lower temperatures. The most commonly known stress studies for Debaryomyces were often conducted under a single temperature to observe the isolated effect of the stressors in question. However, in this study, a series of experiments were developed to understand better the dynamic and assimilation of multiple stressors under different thermal parameters. Due to the wide genomic divergence of the Debaryomycetea group, a genome sequence was performed to identify the phylogenetic space it occupied. The result showed this hansenii specie (strain J6) to be a subspecies of Debaryomyces fabryi. A physiological assessment provided information on population growth and death rate caused by the synergistic effects of the multi stressors conditions.

A menudo, los organismos deben aclimatarse y ajustar su metabolismo para adaptarse mejor a un entorno de constante cambio. Se puede considerar que algunos abientes tienen parámetros normalizados, mientras que otros realmente desafían el estado homeostático de muchos microorganismos; aunque bajo ambos la vida parece prosperar. Este proyecto intentará comprender estas adaptaciones utilizando la levadura marina Debaryomyces hansenii como sistema modelo de estudio. Debaryomyces hansenii es una levadura marina aislada originalmente de un estuario sueco. Se ha demostrado que tolera concentraciones de cloruro de sodio (NaCl) mucho más altas que las que se encuentran en el agua de mar. Esta levadura puede crecer en la presencia de metales pesados, así como en un amplio rango de pH. Además, se sabe que D. hansenii crece en temperaturas que oscilan entre 11 °C y 37 °C (24 °C, óptimo), un rango más amplio que el comúnmente conocido para las levaduras. Se sabe que la temperatura altera la respuesta de tolerancia de D. hansenii hacia otros factores estresantes. Por ejemplo, en combinación con altas temperaturas (~34 °C), el NaCl estimuló el crecimiento más allá del máximo registrado para cualquiera de los factores estresantes individualmente. Además, se observó una mayor tolerancia frente a múltiples factores estresantes a temperaturas más bajas. Los estudios de estrés más comúnmente conocidos para Debaryomyces a menudo se realizaron bajo una sola temperatura para observar el efecto aislado de los factores estresantes en cuestión. Sin embargo, en este estudio se desarrollaron una serie de experimentos para comprender mejor la dinámica y la asimilación de múltiples factores estresantes bajo diferentes parámetros térmicos. Debido a la amplia divergencia genómica del grupo Debaryomycetea, se realizó una secuenciación del genoma para identificar el espacio filogenético que ocupaba. El resultado mostró que esta especie de D. hansenii (cepa J6) era una subespecie de Debaryomyces fabryi. Una evaluación fisiológica proporcionó información sobre el crecimiento de la población y la tasa de mortalidad causada por los efectos sinérgicos de las condiciones de múltiples factores estresantes.
