Gibberellic acid (GA3) and indole-3-acetic acid (IAA) role on Cd<sup>2+</sup> and Cd-QDs uptake by Leucaena leucocephala

At present, there is no regulations for NMs disposal and living organisms are in contact day-to-day to these new small materials. Despite the benefits of nanotechnology, there are issues about the safety and toxicity of NMs that researchers are interested to address. The aim of this research is to compare Cd uptake from Cd<sup>2+</sup> and Cd-QDs sources by Leucaena leucocephala plants, as well as to study the effect of exogenous phytohormones on Cd absorption. In this research, a capping agent thioglycolic acid (TGA) was used for the stabilization of QDs avoiding aggregation. HRTEM image for TGA-capped CdS-QDs showed a spherical morphology with a face-centered cubic structure. XRD patterns confirmed 5 nm QDs crystalline structure according to their characteristic planes. FTIR confirmed the functionalization of QDs with TGA. ICP - OES was used for Cd quantification and to evaluate macro/micronutrients content in L. leucocephala tissues. Physical and chemical parameters analyzed show the effect of Cd speciation inside plants as well as water deficit stress in leaves from plants exposed to 5 ppm Cd<sup>2+</sup> (82 &plusmn; 3 %). Root elongation decreased 35% in plants exposed to Cd2+ + IAA compared to control group. Total dry weight was lower in plants exposed to Cd<sup>2+</sup> (29%) compared to controls. A significant increase in Cd uptake by roots in presence of QDs (1.0 &plusmn; 0.4 mg/g) was observed compared with Cd<sup>2+</sup> uptake (0.15 &plusmn; 0.02 mg/g). A decrease on the uptake of Zn, Mg, K, and Fe in roots was observed in plants exposed to Cd<sup>2+</sup>, but no significant difference was observed in Mg, Ca, and Fe content in roots of plants exposed to QDs treatment. Catalase (CAT) activity in plant leaves exposed to Cd<sup>2+</sup> and phytohormones show an average increase of 97% compared to controls. However, CAT in roots exposed to Cd<sup>2+</sup> decreased 84% compared to controls. CAT activity in plant roots exposed to QDs + IAA increased 195% and 50% in leaves of plants exposed to Cd-QDs + GA3 + IAA. This study will contribute to a better understanding about the effect of the metal chemical speciation on Cd accumulation in plants....
Actualmente, no hay regulaciones para el manejo de los desperdicios generados por el uso NMs, y los organismos vivos se ven expuestos d&iacute;a a d&iacute;a con estos materiales. A pesar de los beneficios de la nanotecnolog&iacute;a, existen problemas sobre la seguridad y la toxicidad de los NMs que los investigadores est&aacute;n interesados en abordar. El objetivo de esta investigaci&oacute;n es comparar la absorci&oacute;n de Cd<sup>2+</sup> y Cd-QDs por la planta Leucaena leucocephala, as&iacute; como estudiar el efecto de la adici&oacute;n ex&oacute;gena de fitohormonas en la absorci&oacute;n de Cd. En este proyecto de investigaci&oacute;n se utiliz&oacute; el agente encapsulante &aacute;cido tioglic&oacute;lico (TGA) para estabilizar los QDs y evitar la aglomeraci&oacute;n. Las im&aacute;genes de HRTEM de los QDs cubiertos con TGA mostraron una morfolog&iacute;a esf&eacute;rica con una estructura c&uacute;bica centrada en caras. El patr&oacute;n de XRD confirm&oacute; la estructura cristalina de los QDs (~ 5 nm) de acuerdo con sus planos caracter&iacute;sticos. La funcionalizaci&oacute;n de los QDs fue confirmada por FTIR. La cuantificaci&oacute;n de Cd y el contenido de macro/micronutrientes en los tejidos de la planta L. leucocephala fue evaluado mediante la t&eacute;cnica de ICP &ndash; OES. Los par&aacute;metros f&iacute;sicos y qu&iacute;micos analizados muestran el efecto de la especiaci&oacute;n de Cd en el interior de las plantas, as&iacute; como el estr&eacute;s por d&eacute;ficit de agua en hojas de plantas expuestas a 5 ppm de Cd<sup>2+</sup> (82 &plusmn; 3 %). La elongaci&oacute;n de la ra&iacute;z disminuy&oacute; un 35% en las plantas expuestas a Cd<sup>2+</sup> + IAA en comparaci&oacute;n con el grupo control. El peso seco total se redujo 29% respecto al control. Las plantas expuestas a los QDs de CdS cubiertos con TGA (1.0 &plusmn; 0.4 mg/g) mostraron un aumento significativo en la absorci&oacute;n de Cd por el sistema de ra&iacute;ces en comparaci&oacute;n con la absorci&oacute;n de Cd<sup>2+</sup> (0.15 &plusmn; 0.02 mg/g). Se observ&oacute; una disminuci&oacute;n en la absorci&oacute;n de Zn, Mg, K y Fe en las ra&iacute;ces de las plantas expuestas a Cd<sup>2+</sup>, pero no hubo diferencias significativas en el contenido de Mg, Ca y Fe en las ra&iacute;ces de las plantas expuestas a Cd-QDs. La actividad de la enzima catalasa (CAT) en las hojas de las plantas expuestas a Cd<sup>2+</sup> + fitohormonas mostraron en promedio un aumento de 97% en comparaci&oacute;n con el control. Sin embargo, las ra&iacute;ces expuestas a Cd<sup>2+</sup> mostraron una disminuci&oacute;n de 84% respecto al grupo control. La actividad de CAT aument&oacute; un 195 % en las ra&iacute;ces de las plantas expuestas a Cd-QDs + IAA y un 50 % en las hojas de las plantas expuestas a Cd-QDs + GA3 + IAA.
