Publication Flavonoid-ferrocene conjugate as new treatment for breast cancer(2021-07-08) Mendoza Morales, William Omar; Meléndez Martínez, Enrique; College of Arts and Sciences - Sciences; Morell Cruz, Luis; Román Velázquez, Félix R.; Department of Chemistry; Plaza Delestre, María L.Research in drug development for breast cancer treatment has been crucial, since traditional treatments have been associated with neurotoxicity and nephrotoxicity. Previous studies showed that when ferrocene is combined with a biomolecule it targets specific cell lines, shows cytotoxic activity and eradicates cancer with high specificity, surpassing traditional treatments. This research focuses on the synthesis and characterization of flavonoid-ferrocene conjugate drugs that can bind and target the overexpressed estrogen receptor, to eliminate breast cancer cells. Structures of the ferrocene chalcone and aurone were determined using 13C NMR and 1H NMR Spectroscopy where two ferrocene-conjugates were synthetized: 1-ferrocenyl-3-(2-hydroxy-5-methyl-3-nitrophenyl)- prop-2-en-1-one and (Z)-5-methyl-3-nitro-2-ferrocenylidiene-benzofuran-3-one. The docking studies of the conjugates were studied on ERα-LBD, 17β-HSD1 enzyme and HSA, using AutoDock Vina to determine the relative score of energy affinity. The data suggest that the ferrocene conjugates could serve as vectors which could aid in the recognition of ERα. In addition, the ferrocene-conjugates showed the capability to enter at the ligand binding site of the of the three proteins in this study. Future projects are in place to apply ferrocene-base drugs to treat different types of cancer cell lines.
